N2 - Fluorescence lifetime imaging (FLI) has emerged as a powerful modality for biochemical and cellular investigations, including in vivo applications; however, visible-range FLI is severely constrained by tissue autofluorescence as well as photon scattering and absorption. To enable deeper tissue imaging, we employ near-infrared-I (NIR-I) excitation, where these effects are substantially reduced. To further overcome the low quantum yield and limited photostability of NIR dyes, we investigate metal-enhanced fluorescence using biocompatible gold nanoparticles conjugated with IRDye 800. Leveraging an advanced time-gated single-photon avalanche diode (SPAD) array camera (SPAD512S), we demonstrate a fast, wide-field approach for robust multiplexed NIR FLI.
